Abstract

General use of antibiotics raises the development of antimicrobial resistance, makes it worth to find a new potent antimicrobial agent. This study aims to provide a novel antibacterial by dimerization of pyrogallol, which has been studied for its antibacterial activity. The reaction between 2,2-diphenyl-1-picrylhydrazyl (DPPH) and pyrogallol succeeded to produce pyrogallol dimer via oxidative coupling reaction. In this research, the dimerization of pyrogallol was carried out through three different methods (previous method of pyrogallol dimer synthesis, antioxidant assay with and without potassium phosphate buffer). Each method’s mixture was analysed using TLC and product of antioxidant assay with potassium phosphate buffer effectively yielded the expected product. LC-MS/MS and 1 H NMR analysis were also executed and the product is confirmed to be pyrogallol dimer. The antibacterial activity of pyrogallol and pyrogallol dimer were evaluated using agar diffusion and broth microdilution. In agar well diffusion, pyrogallol and its dimer showed the same antibacterial activity against S. aureus but the dimer tend to perform higher antibacterial activity against E.coli. Pyrogallol dimer had lower MIC value for both S. aureus and E. coli (8 µg/ml) compared to pyrogallol (512 µg/ml and 256 µg/ml, respectively).
