Abstract
Palm Fatty Acid Distillate (PFAD) is one of the by-products in the refinery process of CPO. It contains bioactive compounds such as Vitamin E, phytosterols, and squalene, which is one of the best natural emollients for pharmaceutical products and cosmetics. Indonesia is the world’s leading producer of CPO and, as the result, also PFAD. The finding of squalene in PFAD could potentially become an alternative source, which not only increasing the use of the CPO by-product and also helping in the environment preservation. In this study, PFAD underwent double saponification process using different concentrations of KOH with the addition of centrifugation process in between and continued with liquid-liquid extraction. The presence of squalene was confirmed using TLC and IR spectroscopy, and then quantified using HPLC. The best experiment variant using 85% w/v KOH conc. yielded up to 14.55mg with purity of 19.5%. The column chromatography showed a tremendous purification capability, increasing the purity of squalene up to 47.7% using n-hexane as the mobile phase and 10g of Silica Gel 60 as the stationary phase.
